That varies, based on the individual contract or source of a magical girl's power, but for the most part, no, physical purity is not a requirement. In most cases, it's spiritual or moral defilement that can invalidate a magical girl's powers- sex, as long as it's done in good-faith (informed consent of all involved, no betrayal of vows, those involved are not known to be a sworn foe of the source of your power, etc), is not going to cause any harm in this regard.  Your best bet is to ask your patron (if said patron is interactive) or to research the source of your powers as carefully as possible and draw your conclusions from what you find out.

You should probably try to ask whoever had the amulet last. Or you could try and find the origin of it. Both of those should let you know if there are anything you should avoid that could possibly cause you to lose your powers. Also, that may not matter if you are someone who is charged with fighting a particular enemy, as you should have assistance in any rules of that kind from the start.

As Aquariousbound suggested, finding out more about the history of your amulet is a good bet. Even if you can't contact the maker or a previous holder, you might be able to get a solid idea of the purpose, rules and trappings of your power based on the reason it was made and how it's been used by others before you.  If that fails, you'll probably have to find a magical expert of some kind- an enchanter, a magesmith, etc- and ask them to study and analysis the magics that make the amulet function. Just be very sure they're trustworthy of course! Another back-up plan is to find a diviner of some kind and see if they can either read the past or just get a few signs and portents to provide a clue.

Ah, the age old issue of defining inclusion. I think the working definition for this thread is something like 'a female that's empowered by magic to uphold certain values or to perform a duty.' So, as a female wizard, I wouldn't technically qualify as a magical girl because my power comes from myself and I'm not obligated/destined/etc to do something in particular with it. Still, that doesn't mean you can't be a 'friend to magical girls.'   :D

That would seem to be a rather good definition, as I am sure there are more stipulations and rules for magical girls as there are for..female wizards? I would usually use magician, as most would think that a female wizard is a witch and that is a very different thing in my experience.

I think that may also be insensitive? Someone I know seemed to imply that some people would view 'witch' as a derogative term for a female magic-user. Also, I thought a transformation was usually required for you to BE a magical girl, even if a guide isn't. Is that not true?

Names and labels are really tricky things, particularly when there isn't a firm authority to decide on things. In my case, I generally go along with the terminology, if not all of the beliefs, of the White Council (the longest running and most numerous group of mortals that cast spells).  In that framework, anyone of a certain level of skill and raw power is a wizard, as opposed to a practitioner- gender is irrelevant to the label applied. Unless of course, you break a Law of Magic in which case you become a warlock. On the other hand, if someone called me a witch, I probably wouldn't be offended if they just meant 'female that casts spells' as opposed to 'female that sold herself to evil for magic.'

Anyway, that's heading off topic... I can see your point about a transformation being somewhat traditional, RockyDiver.  How about...'a female that's empowered by magic, accessed after a transformation, to uphold certain values or to perform a duty' for our definition?
